Principle

This assay employs the quantitative sandwich enzyme immunoassay technique. The micro plate provided in this kit has been pre-coated with an antibody specific to Human IL-1ra/IL-1F3. Standards or samples are added to the appropriate microtiter plate wells then with a biotinconjugated antibody specific to Human IL-1ra/IL-1F3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human IL-1ra/IL-1F3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450nm±3nm. The concentration of IL-1ra/IL- 1F3 in the samples is then determined by comparing the OD of the samples to the standard curve.

Precision

Mean coefficient of variation for Intra-Assay and Inter-Assay: 3 samples with low, middle and high level concentration were tested for repeat multiple times, respectively. The results showed that the coefficient of variation of the kits was less than 10%, which met the precision quality control standard.

Intra-Assay: CV<5%

Inter-Assay: CV<8%
